[Abstract]

In recent years,many tumors suppressor genes have been found which helpus recognize carcinogenesis factors. The role of hMLH1 gene in carcinogenesishas basically attracted considerable attention. Reduction or deletion of hMLH1gene expression in colorectal cancer has been recognized[6,7,10,25], but themechanism which causes the reduction or deletion of hMLH1 gene expressionin colorectal cancer is still unknown. Foreign researches have indicated thatone of important reasons of hMLH1 gene abnormal expression is hMLH1 genepromoter region high methylation[37-39],but the researches like these are very fewin China.Our works is to explore the correlation between hMLH1 gene inactivationand hMLH1 gene promoter region methylation, from which we may find thepathogenesy of colorectal cancer. HMLH1 gene expression was examined withimmunohistochemical method in 40 specimens of colorectal cancer tissue and40 specimens of normal colorectal tissue, and hMLH1 gene promotermethylation was examined with MSP method. The MSP method is the mostextensive technology used to research CpG island methylation, which can beused to research DNA modified by sulfite. Sulfite can change non-methylatedcytosine to uracil, but it can not change methylated cytosine to uracil. Thismethod can cause difference between methylated DNA and non-methylatedDNA, Both full degeneration of DNA and full transition from cytosine to uracilin this method can avoid false positive result. The sensitivity of this technology is so high that it can be used to researchmethylation of small quantity sample DNA, including paraffin imbeddingsample DNA and microdissection DNA. Because of so many superiorities, MSPmethod is considered a fast and high performance clinical research method.HMLH1 gene promoter methylation of colorectal tissue is examined with MSPmethod in the experiment, so that higher detection accuracy can be received.The patients contain 23 men and 17 women, age from 29 to 67, average age 55,neither radiotherapy nor chemotherapy on these patients before their admissionto our hospital in recent two years. The result showed positive rate of hMLH1gene expression was 45.0% in 40 specimens of colorectal cancer tissue, 97.5%in 40 specimens of normal colorectal tissue.
